研究実績の概要 |
This year, the microscope system and imaging analysis software were established for the muscle tissue and muscle stem cell analysis. The immunofluorescent staining methods has been improved and completely established. We found that carnosine-deficient mice exhibited impaired muscle hypertrophy in the cardiotoxin-induced muscle injury model. We further found that carnosine was unlikely to impact on the process of self-renewal, proliferation, and commitment to differentiation, but rater decreases SC susceptibility to early cell death upon activation by applying the single muscle fiber culture technique.

理由
Due to the COVID-19 pandemic, accessibility to laboratories was limited. So, I could not conducted the experiments of effects of carnosine on energy metabolism of skeletal muscles.

今後の研究の推進方策 |
In FY2022, effects of carnosine on energy metabolism of skeletal muscles will be evaluated in CARNS1-KO and WT mice using metabolic cages. Then, effects of carnosine on muscle regeneration ability will be assessed by using CARNS1-KO mice and a cardiotoxin muscle injury (CTX) method. In order to determine if carnosine is involved in proteolysis, muscle-derived mRNA will be prepared to assess gene expressions of proteasome markers MuRF1 and atrogin-1 during muscle regeneration process.
