1. Based on our previous findings, we find Saos2 cells show a better osteogenesis ability on zirconia and titanium than the control group. However, MC3T3-E1 cells could not conduct the osteogenesis ability well on all materials. To detect the reason behind, we introduce a new cell line into consideration human origin MG-63 cells. We seed MG-63 cells at a density of 1x10*4 cells/ml and 2x10*4 cells/ml respectively on the control plate, CpTi, 3Y-TZP and NanoZr surfaces for 7 days. Meanwhile, we detect the ALP activity on Day 7 and Day 14. However, both cell proliferation ability and ALP activity assay did not comes a positive result. Thus, we considerate about the cell ability of MC3T3-E1 cells. MC3T3-E1 cells is the murine calvarial pre-osteoblast cell line and its derivatives are widely used tools in bone research. We pre-treated MC3T3-E1 cells with osteoblast-inducer reagent for culturing and inducing it into osteoblast cells, and find it shows a similar result with Saos2 group.
2. It has been widely known that integrin is the main cell surface adhesion receptor which can detect the exposure of RGD motif of ECM proteins specifically. For detecting the relationship between integrin and Heparan sulphate proteoglycans , we pre-treated the cells with RDG (Arg-Fly-Asp-Ser) and nagstive control reagent RGE (Arg-Fly-Glu-Ser) and want to detect the expression of syndecan family on different materials.
