The aim of this research was to mimic tissue-tissue interactions by combining in a chip device tissues separately cultured in a CUBE device. This was demonstrated with a blood-brain barrier (BBB)-brain model. The BBB module was created by seeding astrocytes and pericytes in Matrigel in the CUBE, with iPSC-derived brain microvascular endothelial cells (BMECs) seeded on the top surface of the gel. The brain module consisted of brain cancer cells encapsulated in Matrigel in the CUBE. Both BBB and brain modules were assembled in a chip device, and a drug for treating brain cancer was applied to the BBB side to simulate the testing of a drug's permeability into the brain. The proof-of-concept showed that tissue-tissue interaction can be mimicked in vitro using the platform developed here.
