This study aimed to develop the proof of concept for sugar-responsive injectable hydrogel and investigate its therapeutic effects on the delivery of stem cells and treatment of myocardial infarction. We successfully synthesized a phenylboronic acid-grafted polymer and achieved a concept of the sorbitol-responsive injectable hydrogel whose sol-gel transition spontaneously occurs via simple diffusion of sorbitol molecule out of the polymeric solution in contact with the heart tissue. The hydrogel formation of our material in the heart tissue was confirmed with no need for external stimulation in vitro and in vivo. In the cellular experiment, we demonstrated that our material has no cytotoxicity. Using a rat model, we showed an intramyocardial injection of our material prevented adverse cardiac remodeling and improved the function post-infarction. Next, we injected a mixture of MSCs and our material into the ischemic myocardium. Following that, the cell viability was tracked by 7T-MRI and fluorescent staining. Those results were published in 2 peer-reviewed papers and five presentations.
