研究概要

本研究において、我々は動脈硬化初期病変のモデルである炎症性刺激によって刺激した内皮細胞における遺伝子発現の誘導と収束のメカニズムを研究した。誘導された転写におけるRNA産生を詳細に観察し、RNAポリメレースII(PolII)の局在変化を経時的に観測したところ、刺激が加わるとmRNA産生に先立って、組織化されたPolIIによる転写が転写開始点から遺伝子終末まで進行していた。この時転写誘導される代表的遺伝子群において、遠隔相互作用が引き起こされていた。ヒストン修飾や主要転写因子の結合部位情報と、全ゲノムクロマチン相互作用解析によってクロマチン構造変化を検討したところ、刺激開始直後から遺伝子間、制御領域間での相互作用がダイナミックに変動している様子が明らかになり、炎症刺激以外でも普遍的な原理であることが示された。
