研究概要

FLASHはヒストンmRNAの成熟や細胞周期S期進行に必須の分子である。本研究では、FLASHがhistone lysine-specific demethylase 1 (LSD1) やcorepressor of REST 1 (CoREST1)と直接結合してCoREST複合体の構成因子となり、エピジェネティックな転写調節に関わることを示した。また、正常二倍体細胞やES細胞ではFLASHはS期進行に必須ではなく、FLASHノックアウトES細胞から神経や心筋、内胚葉やトロホブラストへの分化は正常であった。FLASHはがん細胞特異的に細胞周期進行に必須である可能性が示唆された。
