研究実績の概要 |
In the current study, the applicant (1)successfully fabricated the micropatterned subcellular spatial extracellular matrix (ECM) strips by microcasting method with 1% of DMS-co-EO containing PDMS mold. (2) observation of ECM(collagen) gel strips.(3)the fibroblast cell morphology onto the micropatterned subcellular spatial ECM (collagen) strips was observed. The fibroblast cell elongated its morphology on the collagen strips..
The detailed contents are listed in the following. (1) The applicant has successfully designed a unique substrate with the subcellularsized spatial stripes of collagen ECM strips, It is expected to have different surface mobility. (2) After the collagen strips were fabricated, the applicant did the observation of collagen gel strips by several methods, including florescent staining of collagen gel strips , AFM observation of the 3D topography of collagen gel strips, second-harmonic generation (SHG) observation of fiber structure in the collagen gel strips. (3) Evaluation of the cell morphogenesis on biomaterial surfaces with micropatterned subcellular spatial ECM strips. Cell attachment area and aspect ratio (the ratio of the short axis divided by the long axis of the best-fit ellipse) was assessed.
The significance and importance of the study are that the applicant successfully fabricated substrate with micropatterned ECM strips based on micro-casting. It pointed out the importance of the ECM stirp on the cell morphogenesis, which can work as a fundamental basement for further study of surface mobility.

理由
So far, the applicant has successfully (1) prepared the micropatterned subcellular spatial ECM (collagen) strips (2) fibroblast cell has been seeded on the micropatterned subcellular spatial ECM biomaterial surface and the cell morphogenesis was observed. It can be seen the cell morphogenesishas elongated along the direction of the stripe, which can provide us with evidence that the cell is sensitive to the micropatterned subcellular spatial pattern of ECM. The underlying reason may be that thecell morphogenesishas can be affected by the different surface mobility induced by the micropatterned subcellular spatial pattern of ECM.

今後の研究の推進方策 |
The applicant will continue to fabrciate different micropatterned subcellular spatial pattern of ECM with differnt surface mobility which are based on laminin, fibronectin, and observe the differnt types of cell morphology (not only the current study of fibroblast, but also other types like stem cell) on these substrate with different surface mobility

次年度使用額が生じた理由 |
1. The experiment is in progress, and it is necessary to use the budget to buy some instruments (e.g. cantilevers for AFM), some chemical reagents(fluorescent staining), other types of ECM(laminin) and cell types for the further study. 2. In the last fiscal year, the lab that the applicant worked in is closed, so the plan to buy some instument was delayed. In the next fiscal year, the appicant will work in the new lab and continue with the study. Therefore, the budget of the incurring amount is necessay to be used for the next fiscial year.
