We evaluated the therapeutic potential of endothelial-targeted FoxM1 gene delivery in a mouse model of myocardial infarction (MI). Using nanoparticles conjugated with plasmids driven by the endothelial-specific Cdh5 promoter, we selectively overexpressed FoxM1 in endothelial cells. Echocardiography showed improved cardiac function in FoxM1-treated mice. Immunostaining revealed increased endothelial cell proliferation and capillary density. RT-qPCR showed upregulation of angiogenic and cell cycle-related genes. HE staining demonstrated reduced neutrophil infiltration. These results indicate that FoxM1 promotes endothelial regeneration, reduces inflammation, and enhances cardiac repair after MI.
