研究実績の概要 |
Establishment of a fluorescent-signal based assay for quantifying biodistribution of nanoparticles (anionic liposomes labelled with rhodamine B) in different silkworm tissues following systemic administration. Synthesis and characterization of anionic liposomes loaded with fluorescently labelled siRNA in Tokyo, following protocol provided by collaborator in Germany (University of Wuerzburg). Quantification of RNA knockdown effects by FANA ASOs targeting selected genes (antimicrobial peptide-encoding genes) in primary silkworm cells (hemocytes) in vitro, followed by further evaluation in vivo (silkworms).

理由
During the synthesis and characterization of anionic liposomes loaded with fluorescently labelled siRNA, it turned out that the encapsulation efficiency of siRNA in liposomes varied strongly among different batches, indicating that the protocol for synthesis should be further optimized to reduce batch variations (optimization by collaborator in Germany).

Visualization of interaction between fluorescently labelled nanoparticles (with or without siRNA) and fungal pathogen in the silkworm.
Validating RNA knockdown effects of FANA ASOs on other selected target genes.
Visualization of fluorescently labelled FANA ASOs in silkworm tissues following different administration routes, for example, systemic vs. oral administration, to see how stable FANA ASOs are in silkworms, especially in the latter case.
