研究概要

ヒ素をセファロースに固定したカラムを用いて、ヒ素に親和性の高いタンパク質を調べた。５価のヒ素には選択的に結合するタンパクは無かったものの、３価のヒ素に対していくつかのタンパク質は強い親和性を示した。次に、promyelocytic leukemia (PML)を発現している細胞より可溶性のタンパクを抽出し、ヒ素結合セファロースカラムを用いて、PMLのヒ素への結合能を調べた。PMLのシステインを多く含むRINGフィンガードメインに、ヒ素が３配位で結合するものと考えられた。　ウェスタンブロットや蛍光免疫染色の結果から、ヒ素の曝露によりPMLにSUMO化が起きていることが確認された。
