研究概要

生体脳におけるタウ蛋白を計測する目的で、タウPETプローブ[18F]THK-5105、[18F]THK-5117を開発した。インビトロ結合アッセイやオートラジオグラフィーの結果、これらのプローブはアルツハイマー病のタウ病理像に高い親和性で結合した。そこでこれらのプローブの有用性を確認する目的で、臨床評価を実施した。その結果、アルツハイマー病におけるタウ病理像の好発部位である側頭葉皮質を中心にプローブの高集積を認めた。さらにこれらのプローブの集積は臨床的重症度や脳萎縮と相関した。これらの所見から、[18F]THK-5105、[18F]THK-5117はタウ病理の非侵襲的計測に有用と考えられた。
