研究概要

がんの転移が成立するにはがんが周囲の微小環境にうまく適応する必要がある。
大腸がんは肝臓に高率に転移し、肝転移の制御は大腸がんの治療において最も大きな課題である。われわれはがん細胞自身が分泌する酵素により活性化された肝由来のインスリン様増殖因子(IGF)が肝転移形成に重要な役割を果たし、中和抗体による治療標的となりうることを明らかにした。さらに活性化されたIGFを血中で評価する系を確立し、マウスモデルにおいて転移の形成とともに活性化 IGFが上昇することを証明し、中和抗体治療におけるバイオマーカーとなりうることが示唆された。
