研究概要

１型糖尿病は、膵臓でのインスリン分泌細胞（β細胞）に対して組織特異的自己免疫反応が生じ発症する。β細胞のみが破壊されるメカニズムを解明し治療に応用する目的で、１型糖尿病の候補遺伝子群と複数の臓器特異的自己免疫疾患（β細胞：１型糖尿病、甲状腺：バセドウ病・橋本病、毛包：円形脱毛症）との関連解析を行ない、膵島に対する組織特異的自己免疫反応に関連を示す遺伝因子を解明した。ヒトにおいて関連が示されたMafA遺伝子をノックアウトした１型糖尿病モデルマウスを作出したところ、１型糖尿病発症が著明に抑制され、Mafa遺伝子は１型糖尿病の発症機序に関わる重要な遺伝子であることがin vivoにて確認された。
