研究概要

本邦における死亡の第一原因は癌であり、約半数の日本人がこれに罹患している。そのなかでも、消化器固形癌はその頻度が高く、手術適応とならない進行癌の症例では、有用な治療手段がなく緩和ケアに移行せざるを得ないのが現状である。そこで本研究では、従来の治療法とは全く異なる概念で、アデノウイルスの増殖による遺伝子医薬の有用性を検討し、従来の化学療法剤との併用効果があることを明らかにした。風邪ウイルスである同ウイルスの遺伝子を組換えて、その増殖をヒトの腫瘍で高発現を示す遺伝子の転写調節領域で制御させ、かつ感染効率を増強させた人工ウイルスを作製した。同ウイルスは動物実験でも食道癌に対して有用性を発揮した。
