動く神経細胞の蛍光観察におけるROI設定

研究概要

本研究の目的は、動く線虫の神経活動を定量的に評価するために蛍光画像の動画から蛍光発光強度を調べる領域(ROI:Region Of Interest)を自動設定する手法の開発である。顕微鏡ステージのリアルタイム制御により観察対象の運動を追跡する顕微鏡装置を用いて、多様な刺激に対する生物の反応を多様な観察手法で高い時間・空間分解能で解析・記録する装置を作成した。とくに、多細胞生物で多くの研究にモデル生物として用いられている線虫(C. elegans)を明視野ならびに蛍光観察できるシステムを実現した。さらに、ROIの自動追跡ソフトウェアを開発し、その有効性を確認した。
