細胞増殖・不死化を阻害する低分子化合物スクリーニングと創薬化

研究概要

Baf3 細胞株の増殖・生存を阻害する低分子化合物のスクリーニング→各化合物が標的とする分子の精製・同定→各化合物の臨床応用の可能性の検討、という手順で研究を進めた。約 10000個の低分子化合物をスクリーニングした結果、Baf3 細胞増殖を抑制する 61 個の低分子化合物を選別できた。類似の構造を持つ新たな化合物を加えて、骨髄性白血病細胞株 32D、赤白血病細胞株 MEL などに及ぼす作用を解析するとともに、化合物が水溶性であること、化合物がアミノ基や水酸基をもつこと、などを指標にして、候補化合物を 4 個 (T-167317, T-170633, T-005170, T-051702)に絞り込んだ。T-167317 は、Baf3 細胞特異的に細胞周期を制御した。T-170633 は、Baf3 細胞・32D 細胞・MEL 細胞に対し、細胞の多核化を誘導し細胞周期を制御した。Baf3 細胞・32D 細胞・MEL 細胞に対し、T-0051706 は細胞死を、T-051702 は細胞周期静止を、誘導した。 これら 4 化合物の標的分子の精製・同定をおこなうために、各化合物を FG ビーズに固相化する過程を終了した。
