軸索ガイダンス分子ｄｒａｘｉｎが担う視床皮質軸索投射機構の解明

研究概要

draxinはサブプレート神経を含む大脳新皮質に強く、大脳腹側部と視床に弱く発現する。大脳新皮質に発現するdraxinが視床皮質軸索投射に重要であるかを検証するために、レスキュー実験を行った。pCAGGSプロモーター下にloxpを両端に持つβ-geoとさらに下流にdraxin-IRES-eGFPを持つトランスジェニックマウスを作成した。このマウスを用いてdraxinノックアウトマウスにおいて大脳新皮質特異的にdraxinの発現を誘導した結果、多くの視床皮質軸索が大脳新皮質に向かって投射する事が分かった。
この結果から、サブプレートを含む大脳新皮質に発現するdraxinの重要性が示唆された。
