西南諸島に産する両生類絶滅危惧種の遺伝的多様性の解明と飼育繁殖における遺伝的管理

研究概要

絶滅危惧種の遺伝的多様性を維持することは、その生物種の進化史を後世に残し、さらに将来の環境変動に適応して存続するための進化的可塑性を与える唯一の方策である。本研究では、遺伝的多様性を考慮した持続的な保全と、飼育下繁殖の遺伝的な環境を整備することを目的として、西南諸島に固有な絶滅危惧両生類6種について、マイクロサテライトマーカーの開発と、それらを用いた集団遺伝学的解析を行った。最終年度においては、ハナサキガエル種群におけるマイクロサテライトマーカーの開発を行うとともに、ハナサキガエルの集団構造解析を行い、さらにアマミハナサキガエルにおいて、発生段階における近親交配と温度上昇の影響を検証した。これらの結果は、1報が投稿中、2報が投稿準備中である。また、イボイモリについては、すべての島嶼集団における飼育下繁殖に成功し、国際誌に掲載された。
これにより、本研究の研究期間全体を通じて、最終的に対象種すべてに対してマイクロサテライトマーカーの開発を完了するとともに、集団構造解析を行うことができた。その結果として、島嶼内にも明確な遺伝的集団構造が存在し、それらは進化的背景と、それぞれの種の種生態とそれに関連した景観要因を反映して、独自のパターンを形成していた。これらの結果は、本邦産両生類において詳細な集団構造を解明した初めての研究成果であるとともに、当該種の保全活動への基礎的な情報として重要な知見となり得る。
