研究概要

私達はiPS細胞由来樹状細胞(DC)を作成し，腫瘍抗原遺伝子であるhgp100遺伝子を導入し，これを用いた癌ワクチン療法の可能性をマウスを用いて検討した．結果，iPSDCはnaiveDCと比較して成熟能，遊走能は同等であることを示し，さらにhgp100遺伝子発現iPSDCワクチンによってhgp100特異的な細胞障害性Tリンパ球(CTL)が誘導されること，またこのCTLはhgp100発現細胞株であるB16に対して強力な抗腫瘍効果を発揮することを立証した．また実際の治療モデルでnaiveDCとiPSDCは同等のワクチン効果を発揮することを立証した．今後の臨床応用の可能性が示唆された．
