1,Objective Streptococcus mutans is the primary etiological agent of human dental caries, and forms biofilm on the tooth surface. Many genes associate with biofilm formation of S. mutans. We previously found out SMU832 and SMU833 associated with quorum sensing and biofilm formation in clinical isolates of S. mutans from children and their mothers, using various methods such as DNA microarray. In this study, to clear the roles of these genes to aggregation and biofilm formation, the deficient mutants were constructed and compared with the wild type (S. mutans UA159). 2,Conclusions (1) Gene expressions of SMU832 and SMU833 might associate with aggregation and regulation of biofilm development in early exponential phase. (2) Higher biofilm formation was dependent on the earlier growth in the SMU 832 and 833-deficient mutants. (3) The biofilm formation of the SMU832 and 833-deficient mutants in TSB containing glucose was dependent on extracellular DNA.
