紫外共鳴ラマン分光法によるヘム蛋白質高次構造変化の検出と蛋白質機能制御機構の解明

研究実績の概要

１.ヒトHbのcavity mutant作成とそのリガンド結合による高次構造変化：ヒトHbのα或はβサブユニットのみがFe-His結合を持たない部位特異的変位体を作る事に成功した。すなわち、His-F8をGlyに置換し、His-F8の代りにイミダゾール(Im)をヘムに配位させると、そのヘムにはO2, CO, NO等が結合できるが、ヘムの構造変化がFへリックスに伝搬されない。そのHbの酸素平衡曲線を測定し、リガンド結合による構造変化を1H-NMR、可視／紫外共鳴ラマン、near-UV CD,で調べた。αのFe-His結合を欠くHbは、O2親和性の異なる２つのヘムが存在する２相性の酸素結合曲線を与え、協同性は示さなかった。一方βのFe-His結合を欠くHbは酸素親和性の高いへム１種の存在を示し協同性は示さなかった。分光学的測定結果は、リガンド結合による四次構造変化が起こらない事を示した。αのFe-His結合の消失はHbをTに凍結させるが、βのそれはαのO2親和性を高める事がわかった。
２）可溶性グアニレートシクラーゼ(sGC)モデルとしてのバクテリアHNOXの共鳴ラマンスペクトル：　NOセンサーであるsGCのラマンスペクトルはHbとはかなり異なる。それが蛋白のTyr-Ser-Arg (YxSxR)トリプレットモチーフとヘム側鎖との相互作用のためと想像されている。バクテリアHNOX蛋白がそのモチーフをもち、sGCに似たラマンスペクトルを与える。特異な相互作用を持つ蛋白のヘムの入れ替えは化学的にはできないので、自分でヘムを合成できない大腸菌、RP523株にHNOXを発現させる遺伝子ペクトルを作り、RP523株によるHNOX合成に成功した。それに外部からメソヘムを加えてHNOXが正しくできたので、その共鳴ラマンスペクトルを測定した。
