研究概要

本研究では、歯周細胞の細胞内殺菌機構を解析し、歯周病罹患の遺伝的宿主リスクを判定する予測歯科医学の創生に寄与することを目的とした。
P. gingivalis は歯肉上皮細胞に侵入後，初期エンドソームに存在し、細胞輸送小胞の融合に関わるSNARE タンパク質VAMP2 を介した輸送経路により、細菌の約半数は細胞外に脱出し、細胞内に残った細菌の大半はオートファジー経路に輸送されることが示された。さらに細菌を捕獲するオートファゴソームは小胞体とミトコンドリアの接触点で生成されていることも示された。これは、２つの全く異なるオルガネラが協働して第３のオルガネラを産むというユニークな知見である。
