研究実績の概要

当該年度では、スクリーニングより得られた低分子化合物について、ITCによる熱量測を行い、特異的な結合を示す化合物の選別(ヒットバリデーション)より、CapF、Tob1、DJ-1に対するヒット化合物を得ることに成功した。これらヒット化合物と蛋白質との複合体構造解析結果を、熱量解析と共に議論することにより、１つの蛋白質に対してnMオーダーレベルのリガンドを創出することに成功した。
（１）ヒットバリデーションにおける熱量解析：黄色ブドウ球菌に対する新規抗菌剤開発を目指して、CapF特異的に結合し酵素反応を阻害する低分子化合物を取得するために、東大創薬機構DDIのフラグメントライブラリーを用いることとした。SPRとITCを用いて１つのヒット化合物3-isopropenyl Tropoloneを取得した（KD=13 μM）。得られた共結晶構造解析より、ヒット化合物はCapF内部の亜鉛イオンにキレートし、近辺のcavityに構造相補性をもって相互作用している結合様式が観察された。
（２）蛋白質－蛋白質間相互作用PPIの阻害剤探索：低分子化合物と蛋白質間の相互作用に関する熱量変化に基づく化学的知見を網羅的に集積するため、PPI阻害剤の探索も行った。mRNAの品質管理における疾患に関連するPPIの阻害剤探索として、Tob1-CNOT7をモデルにフラグメントスクリーニングを行った。SPRにてTob1に結合するフラグメントを選出した後、CNOT7との競合アッセイを行い、μM オーダーのフラグメント化合物の取得に成功した。
（３）熱量解析からの高い阻害活性創出　：DJ-1のヒット化合物構造を基本骨格として、DJ-1に対してより親和性が高くグリオキシラーゼ活性を強く阻害する化合物の探索をおこなった。構造情報に基づいた変異体解析、類縁体解析により、DSFとITCによりnMオーダーの親和性・阻害活性をを示す類縁体が得られた。
