What is evolutionary pattern of the domesticated species, it’s wild and weedy relatives? What genomic regions associated with different habitats and contribute to local adaptation in crop wild relatives? What traits are involved? We tackle theses questions by restriction-site-associated DNA (RAD) sequencing technique, morphological and ecological traits. The project is focusing on azuki bean (アズキ), it’s closely wild relatives (ヤブツルアズキ and Vigna nepalensis) and weedy azuki (ノラアズキ). In the first fiscal year, we collected five populations of wild azuki bean (ヤブツルアズキ) and two populations of weedy azuki (ノラアズキ) in southern part of Japan. Habitat and morphological characters, leaf traits, and plant pigmentation were recorded in the field. In China, five populations of the wild azuki bean (ヤブツルアズキ) were collected in collaboration with Chinese researchers. Some seeds of wild and weedy azuki from Osaka Prefecture University did not germinate. Morphological characters including growth habit, leaf and seed traits, flower size and color were measured. DNA extracted from the leaf samples. We also analyzed the patterns of nucleotide polymorphism of the nuclear region. A phylogenetic analysis of our data revealed that the domesticated Azuki bean was related to two genetically distinct wild populations.

今後の研究の推進方策 |
In the second year, the development of SNP (single nucleotide polymorphism) markers using RAD (restriction-site-associated DNA) and morphological characters evaluation are scheduled. Establishing a set of genetic markers is aimed to assess regions of the genome involved in local adaptation and it is important to understand fundamental similarities and differences between populations and divergence of closely species of azuki bean. Once markers are established they can be further studied to look for signatures of selection to any number of evolutionary forces. This step allows us comprehensive study in Azuki. I also plan to extend the collection region to Korea, Nepal, and wide area in Japan. This step helps us for understanding evolutionary history and phylogeographical structure of azuki bean and it's wild relatives.
