The purpose of the present study is to investigate the role of Gremlin in tumor angiogenesis in pituitary adenoma. In GH3, rat pituitary adenoma cell line, double-immunofluorescence analysis we studied with anti-Gremlin antigen and PI. Rat pituitary adenoma cell (GH3) expresses Gremlin. Double fluorescence immunohistochemistry of Gremlin and CD34 localization in pituitary adenoma tissues obtained by transsphenoidal surgery in 45 cases (7 PRL-oma, 17 GH-oma, 17, 2 ACTH-oma, 2 TSH-oma). CD34-positive vessels were coexistent with Gremlin, (Linear regression; p<0.0001 R^2=0.8589). Gremlin and CD34 co-localized significantly, (mean=0. 690). Gremlin and microvascular density (MVD) were detected by double-immunofluorescence microscopy in CD34-positive vessels from tissue microarray analysis in 61 cases with pituitary adenomas (7 PRL-oma, 23 GH-oma, 22 NF-oma, 5 ACTH-oma, and 4 TSH-oma. ). In a tissue microarray analysis, MVD were significantly correlated with increased Gremlin expression, (Linear regression ; p<0.0001 R^2=0.8589). Meanwhile, subtype and Knosp grading were not correlated to Gremlin. In the current study, Gremlin expressed strongly in pituitary adenoma tissue that the tissue has many CD34 positive vessels additionally high coherence of these expression area indicates that Gremlin is associated with angiogensis of pituitary adenoma cell.
