In the last fiscal year, cell wall compositions of soybean and tomato cultivars with different salinity tolerance under various saline conditions were investigated. Two cultivars with higher and lower salinity tolerance of each crop were grown under 0, 40, 80 and 120 mM NaCl conditions. When the growth of the seedlings showed significant differences with different salt concentration, the roots of the seedlings were sampled. Elongation(0-5 mm) and mature (5-10 mm) zones were separately collected. The cell wall composition of pectin, hemicellulose I and II and cellulose were detected. In general, salt treatment decreased the percentage of pectin, but increased that of cellulose across root zones of the cultivars. The tolerant cultivars were able to maintain their main root cell wall substances. The changes in cell wall composition, especially the pectin content showed a close relation with the regulation of root growth. The difference in salt tolerance between the two cultivars of each crop can partly be explained on the changes in response to salinity. Calcium application ameliorated the decrease in the amount of cell wall under salinity stress, especially the pectin fraction. By applying Ca2+, the maintenance of pectin level and increase in cell Ca2+ may contribute to the restoration of root growth under salinity. The ameliorative effect was more conspicuous in the salt-tolerant cultivar than the salt-sensitive cultivar.

理由
The experimental methods for crop cultivation in hydroponic culture and cell wall composition analysis were established. By using these methods, measurements for different crops are expected to be smoothly carried out. Besides, the experimental procedures for investigating root physical properties have also been established in this year. This may step forward the following experiments.

今後の研究の推進方策 |
The future studies will be based on the research plan. Because one plant used in this study is wild species, so the growth of each individual plant differed greatly. By solving this problem, more repetition needed to be added in the experiment, which will take more time for completing the study. A technician will be employed to carry out the repeated experiments.
