Unlike the recovery from anesthesia by most general anesthetics accompanied by the symptoms resembling hangover, the recovery from propofol anesthesia is usually “clear-headed” and pleasant. To confirm the hypothesis of the involvement of the endocannabinoid system in mediating the favorable recovery profile of propofol, the ability of cannabinoid antagonists to deteriorate its quality has to be examined in several behavioral assays of emotional and cognitive functions. These assays are based upon the recording and subsequent evaluation of animal activity, and any interference with animal activity by anticannabinoid drugs will preclude the correct interpretation of the behavioral results. Therefore, in the first part of these study, we employed the open-field test 1) to determine the optimal experimental time schedule that would allow the evaluation of multiple doses of cannabinoid antagonists in the same animal; and 2) to determine the dose range of cannabinoid antagonist that would have no effect on the total distance travelled by mice in the open-field test. These data will be used in the second part of this study to confirm the involvement of endocannabinoid system in positive affective state produced by propofol in the place preference test.

理由

The determination of the dose range of cannabinoid antagonist having no effect on animal activity in the open-field test required more time than planned initially and the testing of the emotional function after propofol could not be performed during the first year of this research project. This is explained by the emerged necessity to determine the optimal experimental time schedule allowing the evaluation of multiple doses of cannabinoid antagonist in the same animal. Such a necessity was unforeseen during the planning phase of this research due to the fact that the principal investigator is new to this kind of behavioral experiments.

今後の研究の推進方策

In the second part of this study we will use the place preference test to confirm the involvement of the endocannabinoid system in positive affective state produced by propofol. To make up for the slight delay in our research, we also plan to examine the effect of the endocannabinoid system modulation on the emotional function during the recovery from non-propofol anesthetics that was initially scheduled for the third part of the study.

The amount for the fiscal year concerned has been almost completely used. The remaining amount (11,275円) is negligible compared to the total amount.

This amount will be used for the purchase of materials (animals or reagents) necessary for the study.
