Initial injections of CNO did not significantly change EEG characteristics in AAV injected and control regions. The pattern of sleep stages also did not change. We repeated the experiments using larger AAV injections, but the results were the same. Measuring the signal correlation between the two recording sites. we found unusually high correlation during waking, NREM and REM sleep and concluded that our surface EEG signals were significantly contaminated by volume-conducted signals, mostly hippocampal theta activity. We changed our recording strategy to ensure that we were recording locally generated signals. We used fine wire electrodes in monopolar and bipolar configurations. Only bipolar electrodes showed truly local signals. Unfortunately, bipolar recordings require twice the number of amplifier connections, increasing the complexity of the surgery and electronics. We have now solved these problems and are repeating the experiments with CNO injections.
