細菌染色体の複製開始と制御の分子機構とその進化に関する研究

研究概要

1.DnaA蛋白とDnaA-boxの普遍性とその進化の解明(吉川担当):大腸菌に加え、枯草菌・Pseudomonus・MicrococcusのdnaA遺伝子の構造を前年度までに決定したが、本年度は、自律的に増殖する細菌として最小のゲノム構造を持つMycoplasmaについてそのクロ-ニングに成功し、DnaA蛋白とDnaA-boxの真正細菌における普遍性を確かなものにした。さらに、下等真核生物である酵母について、相同遺伝子単離の試みを行いその候補となるクロ-ンを得た。DnaA相同蛋白が真核性生物にも存在するか、興味深くかつ重要な問題である。
2.枯草菌DnaA蛋白の機能と発現調節機構の解明(守家担当):前年度までに枯草菌DnaA蛋白とDnaA-boxが複製開始と制御に関与していることを分子遺伝学的に明らかにした。本年度は、精製DnaA蛋白を用いてそのDnaA-boxへの結合様式を解析する一方、抗DnaA抗体を用いてその細胞内の存在量を測定し、ある条件下ではそれが複製開始量を決めることを示した。しかし、dnaA遺伝子の発現を人為的に調節できる系を開発し、染色体複製に対する影響を調べたところ、対数増殖期にある細胞ではDnaA蛋白量の増加が即複製開始につながらず、他の律速因子の存在が示唆された。
3.枯草菌複製装置の構造と機能の解明(小笠原担当):前年度までに枯草菌の第2の開始蛋白DnaBの精製・抗体作製を終了した。本年度は、枯草菌dnaH遺伝子の構造を明らかにし、それが大腸菌dnaZX遺伝子と相同であり、DNAポリメラ-ゼのサブユニットをコ-ドしていることを示した。興味深いことに両遺伝子が共に相同なrec遺伝子とオペロンを形成している点も両細菌で保存されていた。
