研究概要	この研究は、高分解能NMR法を主手段として蛋白質及び蛋白質モデル化合物の種々の環境のもとでの高次構造を決定し、機能との関係を明らかにすることを目的にしており、(1)糖蛋白質、(2)酵素と人工酵素、(3)蛋白質モデル化合物、のそれぞれの高次構造解析と機能の研究を分担課題として昭和62年度から3年間にわたり実施した。最終年度である今年度は、初年度からの溶液状態と固体状態での高次構造と機能についての研究を引続き強力に進めるとともに、3年間にわたる研究を総括した。各課題に関する今年度の研究実績は以下の通り(カッコ内は分担研究者名)。課題(1)ではガラクトサミン基結合スレオニン残基を含むテトラペプチドを不凍性糖蛋白質のモデルとして合成し、糖ーペプチド間相互作用を調べ、昨年度に解析したトリペプチドについての結果と比較、検討した(中条、井上)。課題(2)では化学修飾基としてイミダゾ-ル基を導入したシクロデキストリン(CD)(戸田)と単糖または二糖を枝として導入したCD(山本)の酵素類似機能を調べた。また結合蛋白質プロテインAとメタピロカテ-ゼを融合した蛋白質を遺伝子工学的に合成し発現蛋白質の活性を検討した(相沢)。課題(3)ではポリーγーベンジルーLーグルタメ-トのモデルとして環状2量体を合成し、側鎖の平均コンホメ-ションを決定し(安部)、ポリーLーグルタミン酸塩の溶液中の対イオン凝縮の実験結果を理論と比較した(小見山)。また天然イオノホアのモデルとしてのカルボン酸型ポリエ-テルのアルカリ金属錯体の構造をHーNMR化学シフトから検討した(中浜)。さらに固体状態のトロポミオシンの高次構造を固体高分解能NMRにより検討した(安藤)。
