減数分裂に於ける遺伝子組換え桟構の解析

研究概要	酵母減数分裂細胞特異的遺伝子のクロ-ン化が成功した。このために酵母細胞分裂の詳細な研究と減数分裂誘導条件の改良がすすめられてきた(宮田)。減数分裂細胞由来のpoly(A)^+RNAより合成したcDNAを栄養増殖細胞由来のpoly(A)^+-RNAでサブトラクトしたプロ-ブ(サブトラクトプロ-ブ)を使用し、3回のスクリ-ニングとクロスハイブリッド形成法で相互に独立的14クロ-ンを得た。これらのクロ-ンを用いてノ-ザンハイブリッド法で転写レベルを比較し、14個のクロ-ン全てが減数分裂時に転写量が上昇する単一もしくは複数の転写産物を含むことがわかった。このクロ-ンを用いて調節部分を含む遺伝子全体の解析が進行中である。これらの転写産物の存在に関し充分な注意をもって調査した結果、極めて微量であるが体細胞でも発現していることが発見された。今後その意味が検討される。減数分裂前期の相同染色体対合期とキアズマ形成期にはin vitro系でアッセ-可能な組換え酵素、ATP依存性recA様蛋白(mーrec)、ATP非依存性recA様蛋白(mAiーrec)と3つの関連酵素の存在が示されていたが、他にMg^<++>非依存性蛋白の存在が示された他、シナプトネマ構造(S.C.)の単離と蛋白の精製がすすみ、S.C蛋白もこれらの機構に参加するこが明らかとなった。少くとも5種類の蛋白による保証が存在すると思われる。又組換えに関与したDNA代謝の最終段階ーDNA ligationーについても減数分裂時にはリガ-ゼIの他リガ-ゼIIの活性が高く2重保証システムの存在を示唆した。このような減数分裂前期の特異性は遺伝子の特異性につながるが、酵母で得たクロ-ンの解析とその知見を高等動植物の細胞に応用することで相同染色体の対合とキアズマ形成を分子生物学的に解析する緒につくことができた。
