緑膿菌リポ多糖とくにD-ラムナン結合リポ多糖の構造と生理活性

研究概要

緑膿菌の日和見感染の療法に適したモノクロナル抗体やワクチンの開発に当たっては、比較的広範な緑膿菌に存在する成分がその標的となる。
O-抗原リボ多糖は抗原性は強いが、十数種に及ぶ血清型を示し、免疫療法の標的としては不適当である。他方、微量ながらも広範な緑膿菌に存在する可能性が強いラムノ-ス含有リボ多糖は標的となり得る。しかし、その実在及び構造に関する知見は皆無に等しい。本研究はこれらの点を明らかにし、緑膿菌に対するワクチン・抗体等の開発に役立つ基礎的知見を得ることを目的としている。本研究計画に基づき、以下の結果を得た。
1.本間結成型A,F,K,の三株において、主要抗原の構造を確認した。アルカリ分解に際し、抵抗性の要因となる遊離Co_2H基をメチル化及びアミド化で化学修飾後、アルカリ分解を試行したが、主要抗原の断片化は不完全で、D-ラムナン結合リポ多糖の単離に至らなかった。
2.アルカリ分解法を利用し、本間血清型G,Mのリポ多糖画分からD-ラムナン結合リポ多糖成分の単離に成功した。組成、NMR-スペクトル分析、施光度測定等の結果から、多糖構造を決定した。
3.上記の単離したD-ラムナン結合リポ多糖はモノクロナル抗体E-87と反応することが拮抗酵素結合抗体法により確かめられた。また、その反応性は既報のIID 1008株由来のD-ラムナンと同程度である。
4.D-ラムナンの生合成に関与すると推定されるGDP-D-ラムノ-ス合成酵素は検討した14種の本間血清型標準株の内、9種に存在することが判明した。また、本間血清型における当該酵素お分布はE-87抗体に対する結合のスペクトルと密接に相関することが判明した。
以上の結果から、E-87抗原はD-ラムナン結合リポ多糖であり、この成分は比較的広範な緑膿菌に存在し、共通抗原の性質をもつと推定した。さらに、この抗原に対する抗体は有用な免疫療法剤と期待できる
