腸管神経叢における神経伝達物質の局在と動態。レ-ザ-凝固法と免疫染色法による研究。

研究概要	本年度は、3年継続の本研究年度の最終年度にあたっていた。そこで、昭和62、63年度における研究の成果に基いて、若干の新しい実験を実施し、また研究成果のまとめを行なった。 1.古典的伝達物質と神経ペプチドについて、モルモット腸管神経叢における局在を検討した。 2.レ-ザ-光凝固野の周囲におけるこれらの物質の貯留を研究して、これらの物質の流れの方向を解析した。 3.腸管神経叢のグリア細胞のマ-カ-として、S-100蛋白質を免疫染色したところ、レ-ザ-光凝固野におけるグリア細胞の増殖能が研究できることが判明した。 4.レ-ザ-光凝固直後から、75日経過後まで、継時的にレ-ザ-光凝固野の神経再生現象を研究した。ニュ-ロンの突起とグリア細胞とからなる自律神経基礎網は、グリア細胞の分裂とニュ-ロンの突起の伸縮によって、再生し得るとの結論に達した。正常組織の自律神経基礎網の立体構造と再生した自律神経基礎網の構造を比較検討した結果、正常組織においても、自律神経基礎網は常に改築されているという仮設に至った。 5.自律神経基礎網は、自律神経の終末装置である。その改築現象は自律神経機能の加齢などの理解に重要であると思われたので、BrdU法を応用して、正常組織およびレ-ザ-光凝固野におけるグリア細胞の動態を解析しようと試みた。 6.本研究ではレ-ザ-光凝固法と免疫組織化学法を併用することによってニュ-ロンにおける生理活性物質の流れを研究しようとして開始されたものであったが、結果としてニュ-ロンの中で物質の流れと、神経系の再生・改築現象は密接に関連していることが分かった。
