老年痴呆に関する伝達物質からみた実験的研究ーヒト剖検脳との比較

研究概要

I ラット脳におけるソマトスタチン受容体の局在: 定量的in vitro autoradiography(ARG)を用いて^<125>IーTry^<11>ーsomatostatin(SS)ー14結合部位をラットの前脳と間脳で検索した。大脳皮質ではVーVI層に高密度の、IーIV層に低密度の受容体が見いだされた。嗅内皮質は大脳皮質の中で最も高い受容体密度を示した。嗅系は高いSS受容体密度を示し、前嗅核、外側嗅路核、内側手綱核、底外側扁桃核は中等度の密度を示した。辺縁系では海馬CAI、subiculum、次いでoriens、radiatum、lacunosum層、扁桃核が高い受容体密度を有し、錐体細胞層で低密度であった。尾状核被殼、黒質は低い受容体密度を示し、外障は最高の受容体密度を示した。
II NGF注入ラット脳でのmuscarinic-acetylcholine(mAch)とsomatostatin(SS)のreceptorとの関係に関する実験的研究:mAchR結合部位に対するSSの効果をnerve growth factor(NGF)注入動物において検索した生後2ー8日のラット仔に5μgのNGFを脳室内投与し、12日後に海馬をとりだした。SS投与を受けていない又は受けた対照群と、SS投与を受けていない又は受けたNGF投与群からなる4群に分け、各群でoxotremorine/^3H-N-methyl-scoporamine(^3H-NMS)阻害実験を行なった。脳室内MGF投与動物では海馬のP_2分画を用いた阻害実験はK_i値0.46×10^<-6>Mの単一結合部位を示し、海馬のmAchR結合に対するNGF脳室内投与の効果はSSのin vitro投与の効果と同様であった。NGF注入動物の海馬ホモジネ-トに対するSS添加はoxotremorine/^3H-NMS阻害曲線に影響を与えなかった。
我々は昨年fimbria-fornicotomy後のbasal forebrainのコリン作動性ニュ-ロン数の減少がNGF注入動物では対照に比して軽度に留まる傾向があることを報告した。今回のmAchとSS receptorの関係についてのoxotremorine/^3H-NMS阻害実験からはNGFの影響を示唆する成績は得られなかったが、今後もNGFの生物学的作用の研究を継続する予定である
