酵母のミトコンドリアのリボゾ-ム形成における核とミトコンドリアの協調

研究概要

本研究はパン酵母(Saccharomuces cerevisiae)の細胞における核とミトコンドリアの遺伝情報の関わり合いの一端を解明するために、全ての細胞生物に普遍的に存在するリボソ-ムを対象として、その生合成における核とミトコンドリアの遺伝的協調機構を探ることを目的とする。酵母は最も体制が簡単な単細胞の真核生物であり、遺伝学的、生化学的、分子生物学的な多くの研究の蓄積がある。そこで本研究では、(1)まずミトコンドリアのリボソ-ム蛋白質を精製してアミノ酸配列を決定し、得られたデ-タに基づいて対応する遺伝子のクロ-ニングと構造解析を行ない、(2)ミトコンドリアの全リボソ-ム蛋白質に対する抗体を作製し、免疫学的検出法によって対応する遺伝子のクロ-ニングと構造解析を行ない、さらに、(3)酵母の染色体DNAの整列クロ-ンを作製し、酵母の染色体の構造的特徴を探るとともに、ミトコンドリアの進化の過程で、始原ミトコンドリアから核に移行したと考えられる遺伝子の同定と解析を行なった。その結果本年までの研究でミトコンドリアのリボソ-ム蛋白質の遺伝子を20遺伝子クロ-ンして構造解析を行なうとともに、酵母の第I,III,V,VI,VIIIの5本の染色体の整列クロ-ンバンクを完成あるいはほぼ完成することができた。これらの研究成果の一部はすでに論文にまとめて報告し、また他の一部も論文としてまとめつつある。
今後は本研究の成果の上に立って、ミトコンドリアのリボソ-ム蛋白質を指標として、核とミトコンドリアの遺伝情報がどのような相互作用を行なっているかという点について、当初の目的とした線に沿って研究を継続していきたい。
