ムスカリン性アセチルコリン受容体の構造と機能及び機能調節

研究概要

(1)アゴニスト依存性にムスカリン受容体をリン酸化する酵素(以後受容体キナ-ゼ)を部分精製した。大脳、心房いずれから精製したムスカリン受容体もカルバミルコリン存在下にリン酸化された。ムスカリン受容体をG蛋白質(GO)と再構成させてから受容体キナ-ゼと反応させると、アゴニストの有無にかかわらずムスカリン受容体のリン酸化は見られなかった。そこにGTPγS(あるいはGTPかGDP)を加えると、またアゴニスト依存性にリン酸化が見られるようになった。ムスカリン受容体へのアゴニスト結合によりリン酸化部位が露出されるが、アゴニスト・ムスカリン受容体・G蛋白質複合体の形成により隠され、GTPあるいはGDPの添加で複合体が解離すると再度現れると、解釈される。この結果は、アゴニスト結合及びグアニンヌクレオチド結合の熱力学的あるいは速度論的解析の結果と照合する。(2)培養細胞(NG108 Neuroblastoma-Glioma HybridomaとChinese Hamster Ovary Cells)にムスカリン受容体mlまたはm2サブタイプを発現させ、アゴニスト存在下での受容体のリン酸化を測定する実験を行なった。しかし受容体のリン酸化は観察できなかった。受容体発現量の増加、[^<32>P]リン酸の増量、反応後の受容体回収の効率化などの条件検討を更に続ける必要がある。(3)心房から精製したムスカリン受容体(m2サブタイプ)も試験管内再構成系で3種のG蛋白質(Gi,Go,Gn)と相互作用した。精製受容体の代わりに心房膜標品を用いて、ムスカリン受容体とG蛋白質の相互作用を見る系を開発した。受容体とG蛋白質の相互作用が見られるためには0.3%程度のコ-ル酸存在下で両者を混ぜることが必要であった。この条件でも膜中のムスカリン受容体と3種のG蛋白質との相互作用が観察された。受容体とG蛋白質の相互作用を特異的に保つなんらかの機構があるとしたら、その機構は低濃度のコ-ル酸処理で失活するような不安定なものと考えられる。
