予定外胚葉細胞に対する中胚葉性誘導物質の作用と遺伝子発現

研究概要	両生類の予定外胚葉細胞は末分化細胞として用いられている。末受精卵から原腸胚期の予定外胚葉細胞の細胞小器官の変化が電顕を用いて、形態学的に詳細に調べられた。その結果、卵黄粒の崩壊は排卵時からすでにみられ、桑実胚頃に著しくなる。またミトコンドリアは発生過程を通じて量的変化はあまりみられないが桑実胚期以前では未成熟型であり、それ以降は成熟型のミトコンドリアとなる。またr-ERは、桑実胚期頃から発生にともなって量的に増加してくることが明らかとなった。一方、中胚葉性分化因子の精製・単離についても行った。K-562株やTHP-l株、あるいは繊維芽腫瘍由来のHT・1080など22種の細胞株のコンディションメディウムの中に中胚葉性分化誘導能をもつ物質が存在するかどうかをイモリ胚およびツメガエルの予定外胚葉を用いて行われた。この中から9種の細胞株のコンディションメディウム中に高い中胚葉性の分化誘導能があることがわかった。これらの物質の分化検査方法は予定外胚葉を用いてのサンドウィッチ法やピ-ス培養法がとられた。DEAEカラムクロマトグラフィ-や逆相カラムクロマトグラフィ-などで分析していくと単一の物質が精製された。その後、アミノ酸配列等を調べてみるとEDFと同一の物質であった。これらは現在、またアクチビンAともよばれる物質である。そこでクロ-ニングした遺伝子より合成したアクチビンA(またはEDF)そのものを用いて誘導能を調べたところ10ng/mlでも約70%が中胚葉分化を示した。そして50ng/mlでは調べた外植体のほぼすべてに筋肉や脊索、間充織など中胚葉性分化がみられた。このことはアクチビンAは中胚葉分化誘導因子の一つであり、これは現在まで知られている因子の中で最も低濃度で中胚葉分化誘導をおこす物質である。
