Mycosphaerella属植物病原菌の宿主特異性決定因子に関する研究

研究概要	前年度に引きつずき、本年もエンドウ褐紋病菌(My.pinodes)柄胞子発芽液中に生産される宿主特異性決定因子の精製を試みた。大量に回収した発芽液低分子画からAmberlite XAD-8で活性部を濃縮し、さらに分子ふるいHPLC(YMC-diol,H_2O,1ml/min)により分画した結果、保持時間14.5、17、29分に活性が認められた。大量分取にはHW-40FおよびBiogel-P-4を用いたが、後者で良好な分離が認められた。この結果、2成分に顕著な活性が存在し、これらの分子量は約3500-5000と推定された。現在、機器分析を行っているが、糖鎖(10<)を有するペプチドであることが判明している。キク花腐病菌及びウリ類蔓枯病菌の宿主特異性決定因子の検索を行った結果、いずれも菌の柄胞子発芽液にもそれぞれの宿主に特異的な感染誘導活性を示す低分子物質が見いだされた。即ち、既報の方法に従って低分子分画を回収し、これをキクやキウリに全く病原性のないエンドウ褐紋病菌に混合してキク花弁やキウリ子葉に接種した結果、それぞれの病原菌から得た低分子画分を添加した場合のみ、褐紋病菌の感染の成立と病斑の形成が認められた。水対照区褐紋病菌の因子ではこのような効果はなかった。現在、これらの物質についても分離精製を進めているが、本研究で構造決定に至らなかった最も大きい理由は、本物質の性質にあるものと考えられる。本研究の過程で、褐紋病菌の宿主特異性決定因子について種々の角度から生物活性を調べた結果、本因子は単にファイトアレキシン生合成の抑制作用を有するのみならず、宿主エンドウのATPaseを阻害する活性を示すこことが判明した。本結果は、病原菌は宿主植物の膜系に存在するイオン輸送やエネルギ-生産に関与する基本的な代謝系を特異的に阻害することによって宿主の抵抗性の発現を回避し、感染を果たすことが強く示唆された。
