研究概要	今年度の研究実績は1.GTP結合蛋白質(以下G蛋白質)研究の新しい展開、2.細胞増殖の機構、3.諸種G蛋白質、の3つの分野に分れる。1.に関しては、(1)G蛋白質の進化について粘菌、線虫にIAP基質G蛋白質の存在を証明した(2)酵母のgst1変異を相補する遺伝子のホモローグがヒトにも存在し、ある種のG蛋白質を指令する。(3)細胞接着レセプター刺激で発現する細胞間相互作用や好中球の貪食がIAPで抑えられることからG蛋白質の関与が推測された。(4)HLー60細胞の分化はINFγやTNFで弱く、ビタミンAで強く誘導される。低濃度のビタミンAで短時間培養してもほとんど分化しないが、このビタミン処理細胞にINFγを添加すると著しく分化し、このINFの作用が特異的にG蛋白質を介する。(5)3T3線維芽細胞では、ブラジキニン受容体がGiー2を介してホスホリパーゼA_2を直接的に活性化する。2.に関しては(1)DNAプライマーゼ〜DNAポリメラーゼα複合体を解離・再構成し、プライマーからDNAへの合成の移行の機構を解析した。(2)マウスFM3A細胞から3種のDNAヘリカーゼを精製した。(3)トポイソメラーゼIIがリン酸化によって活性化された。(4)ヒストンH2Aのユビキチン化がDNA合成に必須である。(5)低濃度血清存在下、EGFやホルボールエステル単独で増殖する細胞株を分離し、G蛋白質やCキナーゼの役割を見出した。3.については(1)これまで1種といわれていたGoをさらに3つのサブタイプに分離精製した。(2)通常はコレラ毒素の基質とはならないGiがfMLP受容体を介して活性化された時に限ってこの毒素でADPリボシル化される。すなわちコンフォーメーション変化が推測される。(3)ボツリヌス毒素の産生するC3によるrho蛋白質のADPリボシル化を許容する蛋白質を発見した。(4)低分子量G蛋白質の活性化を促進する新しい蛋白質を発見した。(5)疎水性ペプチドがG蛋白質を直接活性化した。受容体蛋白質のモデルである。
