研究概要

(1)クモ毒素利用によるグルタミン酸電流の解析:グルタミン酸レセプタ-に阻害活性をもつクモ毒素およびそのアナログ化合物を用い、海馬スライス標本および培養海馬細胞におけるグルタミン酸電流に対する作用をしらべた。この結果クモ毒素が非NMDA型レセプタ-チャンネル電流を阻害しNMDA型レセプタ-による電流と区別されることを明らかにした。
(2)脳グルタミン酸レセプタ-分布の組織化学的研究:ビオチンを結合させたクモ毒素JSTXを用いアビジン・ビオチン反応を利用することによりラット小脳および海馬におけるグルタミン酸レセプタ-の分布を明らかにした。小脳では分子層とくにプルキンエ細胞の樹状突起に強い結合反応が見られた。この結果は平行繊維とプルキンエ細胞の樹状突起のシナプスがキスカル酸型であることと対応する。
(3)グルタミン酸レセプタ-の精製:哺乳動物脳から海馬及び小脳のシナプス膜分画を作成しTriton Xー100で可溶化しJSTXをリガンドとするアフィニテイ精製を行い、クモ毒素と特異的に結合する蛋白が得られた。SDS電気泳動法によりこの蛋白は分子量は4万から13万の範囲に数個存在することが判明した。精製した蛋白を人工脂質膜に再構成しパッチクランプ法で電流計測を行った結果グルタミン酸により活性化されクモ毒素により阻害される単一チャンネル電流が得られた。
(4)虚血後のスナネズミの海馬によりスライス標本を作成し、血流停止後のさまざまな時間経過でCA1錐体細胞に起こる膜電位変化を記録し、遅延性神経細胞壊死の過程で起こるグルタミン酸放出、カルシウム代謝異常を裏付ける電位変化を観察した。
