研究概要

大腸菌は遺伝学的に最も詳細に解明されてきた生物であり、現在その全ゲノムの約25%の塩基配列が決定されている。しかし、これまでに塩基配列決定のなされたほとんどの部位は遺伝子およびその近傍の領域であり、遺伝子間の構造的特徴やゲノムの最小構成などについてはまだ不明の点が多い。そこで本研究は昭和62年度に発表されたKohara,Akiyama&Isono(Cell 50,495-508,1987)の大腸菌の「整列クロ-ンバンク」を活用し、大腸菌の全ゲノム構造を解明することを究極の目的として企画された。本研究ではこの目標を達成するための種々の方法や技術を検討し、同時にそれらを使って実際にゲノム構造の解明に向けての解析を行ない、さらにこの目標を達成するためには不可欠とえられる共同研究体制のあり方を検討した。また、「大腸菌のデ-タベ-ス」を作成し、国内外の研究者およびそれらの研究者の研究動向を把握すること、およびより高効率なDNAの塩基配列決定法を確立すること、などに的を絞って検討を加えた。
本年度から重点領域研究「大腸菌のゲノム全構造の解析」が発足したため、本研究はこの重点研究と重複する形となったが、重点領域研究の総括班と協力して上述した諸点の実現を図った。
本研究および上述の重点領域研究により、遺伝子の存在の有無とは無関係に構造解析が行われつつあるので、今後、(1)そのような領域にも遺伝子が存在するのか、(2)存在するとすればどのような遺伝子なのか、(3)大腸菌のDNA中に機能をもたない「無意味な」領域があるのか、(4)ゲノムの構造を特徴づけるものは何か、などの「ゲノム解析」を行なっていくことが可能となるであろう。
