植物の系統形成における代謝期染色体の核型論的研究

研究概要

1.代謝期染色体の形態学的観察について。
シダ植物全般、裸子植物針葉樹類、被子植物バラ科、キク科、ユリ科、ラン科について酢酸オルセイン染色法を用いて、各種群の代謝期染色体の観察を行った。その結果、各種群に対応した形態的特徴がわかった。
(1)代謝期の核型と系統群との間では或る範囲(連、科、亜科、属など)で一致が見られ、系統進化を或る程度反映していることがわかった。
(2)代謝期の核型と地理的分化、生態的分化との間にも関係が見られることが予測された。
(3)代謝期の核型と中期染色体長との間に相関関係が認められた。
2.形態の定量的分析について
代謝期染色体を定量化するために画像解析法を用いることが有効であり、画像作成条件としてネオパンFとミクロファインの組み合わせが優れていることがわかった。
3.細胞学的分析について
ラン科植物31種について間期核クロマチンの形状と核内分布および核DNA量と間期核型との関連を調べた結果、前染色体型はDNA量が少なく、分散型がDNA量が多い、染色中央粒型はその中間のDNA量を示した。また、ネギにおいて、その代謝期凝縮塊を形成するDNAが380bpを単位とする反復DNAであることがわかった。
その他、染色質の質的違い、染色体の分節構造との関係、クロマチン微細構造の違いなどが代謝期核の形態形成に関連あることが解明されつつある。
