研究概要

前年度の成果を基盤として、神経内分泌系を含む広義の泌尿生殖内分泌系の経時的変化とそれらの変化を誘起する機構について、形態学的、内分泌学的、生理学的、薬理学的および分子生物学的手法を用いて解析し、以下に箇条書きする成果を挙げた。
1.生殖内分秘機能を司る視束前野-視床下部および大脳辺縁系の発生に対するステロイドホルモンと神経成長因子の作用を調べ、それらが脳の機能を調節する重要な因子であることを確めた。
2.神経内分泌調節は、液性のフィ-ドバック機構だけでなく、生殖腺と脳との間の神経連絡が重要であることも明らかになった。
3.エイジングに伴うラット血中のプロラクチン増加に注目し、老若ラットの下垂体細胞の培養下でのド-パミンやド-パミン前駆物質に対する反応性を調べ、明瞭なエイジ差の存在を証明した。
4.下垂体プロラクチンの単一細胞レベルでの分泌能の解析に逆転溶血斑法を確立し、ド-パミンの御制作用を発現させることができた。なお、ド-パミン機能が老齢ラットで低下することがプロラクチン分泌増加の主要な原因の1つであることはすでに知られている。
5.老齢ラットにおけるプロラクチン産生下垂体腫瘍はエストロゲン誘発若齢下垂体腫瘍に比べ、プロラクチンmRNA量は明らかに低かった。
6.中年で多飲多尿を示すウイスタ/TW系ラットにおいてエイジングに伴う水代謝、クレアチニン代謝、オキシトシン、バソプレシンの変化を調べ、明瞭な指標を多数確立した。
7.ラットのエイジングに伴う成長ホルモンの低下が、成長ホルモンmRNAの低下と密接な関連のあることも明らかになった。
本年度はこの総合研究の最終年にあたるので、2年間の成果をまとめた研究成果報告書を作成した。
