軸索輸送の基礎と臨床の分子生理学的研究

研究概要

神経軸索輸送のメカニズム、生理的意義そしてその病気との関連について基礎医学と臨床医学の両面から研究討論を行った。広川らは順行性軸索流のモ-タ-分子の候補であるkinesinの分子構造が80mm長の杵状分子であり、一端に10mm径の球状頭部二つ、又他端は扇状を呈する事を明らかにした。抗体修飾法により球状頭部は垂鎖で形成され、扇状端に軽鎖が存在し、頭部で微小管に結合する事を明らかにした。また初代培養神経細胞に蛍光標識チェブリン及アクチンを微量注入しその後レ-ザ-光照射による蛍光退色回復法を行う事により、チュブリン及アクチン等がオリゴマ-の形で送られ神経軸索中で重合体に組み込まれてタ-ンオ-バすることを見出した。小管は有機溶剤中毒の中心をなすn-ヘキサン中毒ラットの坐骨神経知覚線維ではニュ-ロフィラメント蛋白の輸送のみが特異的に加速されているのを見出した。竹中らは軸索輸送に及ぼす神経化学伝達物質の効果を研究した。上頸神経節細胞の軸索輸送はアセチルコリンで可逆的に抑制されることを見出した。また脊髄後根神経節細胞もGABAで同様の抑制効果が見られた。この生理学的意義は極めて重要であり、これからの研究が期待できる。天野は抗ド-パミン作用をもつクロルプロマジン、ハロペリド-ル等の無精神薬がラットの咬筋運動ニュ-ロンのHRP逆行性軸索輸送を促進することを明らかにした。病理の面で藤沢は脊髄神経節について、その軸索部の遠位部の変化と細胞体核周辺部の変化とを比較検討した結果、病変は前者により早期により顕著に出現する事を確認した。千原は筋肉のジストロフィ-のモデルmdxマウスを用い、その座骨神経内のアセチルコリンエステラ-ゼの軸索内輸送について調べ、座骨神経結紮物質の近位にアセチルコリンエステラ-ゼが正常より多くたまることを見出した。
