細胞分裂酵素・蛋白質系の解析と再構成

研究概要	1.大腸菌の細胞分裂・生長に関与するmra遺伝子群の解析。大腸菌染色体地図上2分に位置する十数キロベ-スのmra領域の完全な遺伝子地図の作成とDNA塩基配列決定を行った。この領域に属する遺伝子のうち、細胞分裂遺伝子として同定されたftsWは、その遺伝子産物であるFtsW蛋白質の一次構造が大腸菌の形態形成に関与するRodA蛋白質、枯草菌の胞子形成に関与するSpoVE蛋白質と極めて類似しており、これらが夫々のペニシリン結合蛋白質と対応して細胞分裂、形態形成及び胞子形成を行っていることが予測された。また大腸菌染色体地図上71分に位置するmre領域の遺伝子群mreB、C、Dを同定し、DNA塩基配列決定を行った。これらの遺伝子群はみなそれぞれ細胞分裂を抑制し、細胞の長軸方向への伸長に働くことを明らかにし、細胞の分裂と生長をスイッチオン・オフする極めて重要な機能をもつことを示唆した。 2.黄色ブドウ球菌のβーラクタム高度耐性菌(メチシリン耐性ブドウ球菌)の耐性原因と考えられるペニシリン結合蛋白質の遺伝子を感受性黄色ブドウ球菌に導入してβーラクタム高度耐性にすることに成功し、本ペニシリン結合蛋白質が黄色ブドウ球菌のβーラクタム高度耐性の原因であることを明らかにした。 3.マウスの半数体未受精卵を発生させ、8細胞期で通常の2倍体細胞と集合させることにより半数体細胞由来の細胞を含むキメラ個体まで発生させたが、九日胚以降の段階で半数体細胞はすべて倍数化して2倍体細胞になっていた。しかしそれ以前で半数体細胞は胎児及び胎盤等のほとんどあらゆる所に分化し得ることがわかった。
