細胞分裂酵素・蛋白質系の解析と再構成

研究概要

最も単純な細胞に於て、生物の基本的な営みである細胞分裂の機構を解明し、細胞増殖の系を再構成することを目的として、以下の研究を行った。
1。大腸菌の細胞生長と分裂に関与する<mra>___ー遺伝子群の解析。大腸菌染色体地図上2分に位置する20キロベ-ス<mra>___ー領域の完全な遺伝子解析をほヾ終了した。<mra>___ー領域の5^1ー末端に近い所に存在する330塩基対のオ-プンリ-ディングフレ-ム中に細胞生長と分裂のそれぞれを欠損させる変異があることを明らかにし、また<mraY>___ー、<murG>___ー遺伝子がペプチドグリカン生合成におけるリピド中間体生成の2つの酵素をコ-ドすることを明らかにした。
2。大腸菌の細胞生長と分裂を制御する<mre>___ー領域(71分)に属する<mreB>___ー、<mreC>___ー、<mreD>___ー遺伝子の上記の制御機能を同定し、また<mreD>___ー遺伝子の下流(3^1ー末端の外側)にあるorfE、orfFの塩基配列を決定し、またそれらの機能を推定した。
3。黄色ブドウ球菌のβーラクタム高度耐性菌(MRSAと略称され、病院感染の大きな問題になっている)の耐性原因と考えられる遺伝子、<mecA>___ーと、その発現を制御すると考えられる<mecB>___ー遺伝子の解析を1960年以降に世界で分離されている様々なMRSA株について行った。
4。マウスの半数体細胞系列を分離する目的で,半数体未受精卵を発生させ、これより遺伝子導入其他の方法で安定に自己増殖する半数体培養細胞を育成することを試み、有用なデ-タ-を得た。
5。細胞骨格系蛋白質乃至細胞骨格様蛋白質の分離と解析をすゝめた。
