色素体の分裂装置に関する免疫電子顕微鏡法による研究

研究概要	原色素体がDNAの均等な分配をともないながら分裂を繰り返し、体積を増大させて、その機能を十分に発揮できるように発達したのが葉緑体をはじめとする種々の色素体であると言える。1986年に色素体キネシスの研究過程で、分裂中の色素体のくびれの部分に、色素体の分裂装置ともいうべきリング構造を発見し、"色素体分裂リング"と名付けた。本年度の研究の目的は、単細胞紅藻Cyanidium caldariumから高等植物に至る代表的な植物を材料として用いて調べてきた結果を整理し、この分裂リングの機能、超微細構造及びその構成物質についての全体像を系統を含めて明らかにすることである。平成3年度はこれまでの研究成果を踏まえ次のように進めた。 1.抗アクチン抗体及びその他いくつかの抗体を使ったコロイドー金免疫電子顕微鏡を用いて、分裂リングの形成とアクチン分子の動きとの相関関係を明らかにした。2.単細胞のクラミドモナスのような真核生物からシダに至るまでのいくつかの標本を選出し、分裂リングの観察を行った。3.アラビドプシスの植物体とタバコの培養細胞について色素体の分裂時期を同定し分裂リングの存在を明らかにした。4.タバコの培養細胞もしくは分裂リングが明瞭に現れる材料をもちいて、各種遠心法を駆使して、分裂期にある色素体をプロトプラストから単離する方法を開発を行なった。5.次に、色素体の分裂リングを形成している時期にある色素体だけをより純化する免疫的方法を開発した。6.これまでの結果を体制の簡単な藻類から高等植物に至るまで系統的に整理し、色素体の分裂の一般性を明らかにした。7.純化した分裂期色素体を種々の界面活性剤で処理して、色素体全体を可溶化し、分裂リングを分離することを試みたが分裂リングの化学組成を生化学的に調べるに至らなかった。
