卵外被物質の産生細胞の特定と受精における役割

研究概要

卵外被物質の産生細胞の特定と受精における役割に関する研究(助成期間、昭和63年度〜平成元年度)を進め、平成元年度は次のような研究実績を得た。1)前年度精製した卵外被物質中の精子先体反応誘起に係わる高分子、フコ-ス硫酸タンパク質複合体およびその構成成分(糖鎖結合タンパク質部分、構成タンパク質、糖鎖部分)個々に対するポリクロ-ナル抗体を作製し、前年度採集し、固定しておいた卵巣(10月から次の年の1月末までの期間)の3μ切片を酵素抗体法を用いて免疫組織化学的に検討した。その結果、フコ-ス硫酸タンパク質複合体に対して陽性反応を示す物質は未成熟な卵母細胞内に出現し、卵母細胞の成熟に伴って卵外に出現することが明らかになった。このことは、ウニ卵外被に存在する先体反応誘起に係わる主物質は卵母細胞内で合成され、卵母細胞の成熟に伴って卵外被に分泌されることを強く示唆するものと考えられる。卵巣の湿重量当りのタンパク質およびフコ-ス量は卵巣の成熟過程を通してほヾ一定に保たれていることから、精子先体反応に関与する特異的物質が卵母細胞の成熟過程の特定の時期に合成され、特定の部位(卵外被)に蓄積されることを意味するものと考えられる。2)卵外被に存在する精子先体反応に関与する主物質はフコ-ス量に対してほぼ2倍量のタンパク質を含む推定分子量100万の高分子であるが、この複合体は分子量約26万、24万、12万のタンパク質と分子量約40万の糖鎖結合コアタンパク質とから構成されていると思われる。これらの構成タンパク質はジスルフィド結合を介して会合されていることを示唆する結果を得ている。さらに、分子量約12万のタンパク質が複合体の形成に重要な役割を担っていることを示唆する結果も得ている。
