研究概要

1)吸血に伴って発達し始めた卵母細胞は吸血開始後4日目に卵巣表層から血体腔に突出し、卵巣上皮細胞由来の索状細胞によって支えられるようになった。卵黄形成と卵殼合成は飽血離脱日に開始された。卵黄形成期には、多数の電子密度の高い顆粒が細胞質表層に集積し、よく発達した微絨毛と活発な飲作用が形質膜に認められた。成熟した卵母細胞は索状細胞間隙を通過し卵巣腔へと排卵された。多数の精子が排卵日に卵巣腔に達し、それらの幾つかは卵巣上皮細胞の微絨毛と接し、あるいは索状細胞形質膜の嵌入部に侵入していた。排卵された卵にはもはや特徴的な大きな卵核が認められなかったので、精子は卵巣腔において卵母細胞を受精させると推察された。交尾によって雌性生殖道内に搬入された精子による受精のタイミング、受精場所及び受精のメカニズムについては目下鋭意、追究中である。2)フタトゲチマダニ若虫における宿主血液消化を飽血離脱から脱皮後5日目まで調べた。中腸に含まれる総蛋白質量は離脱から脱皮直前まで急激に減少し、その後の減少は非常に緩慢であった。中腸に含まれる蛋白質の大部分はヘモグロビンとアルブミンであり、これらの消化速度は前者で後者よりも速かった。ダニの中腸に含まれる消化酵素はin vitroの実験からアルブミンよりもへモグロビンを速く分解することが知られ、本実験において認められた消化速度の差異は、カテプシンDと思われる酵素の基質特異性に起因すると推定された。ヘモグロビン消化の残余物ヘマチンはヘモグロビンの減少につれて増加した。ヘマチンは脱皮後1〜5日目に急激に排泄された。更に、宿主血液特にヘモグロビンは、吸血中及び飽血離脱後における生理的に異なった諸相に同調して出現する新生中腸上皮細胞により細胞内消化され、ヘマチンとして排泄されることが電顕的にも裏付けられた。
