研究概要	生体防御の最前線の1つである皮膚からリンパ節を経由して展開される免疫応答のしくみのなかでランゲルハンス細胞(L細胞)は抗原提示機能を発揮するとみられている。本研究はこの細胞の発生と細胞動態を調べることで、皮膚関連免疫系の理解を深めようとするものである。昭和63年度および平成元年度は、L細胞の機能的マ-カ-であるIa抗原に対する免疫細胞化学的方法、ATPaseをマ-カ-とする細胞化学的方法、および電顕的方法を用いて、皮膚、関連リンパ節、皮膚類似構造を示す口腔・消化管上部粘膜に出現するL細胞の検索を行なった。また、本年度はL細胞とThyー1陽性細胞の相互関係およびトリの表皮におけるATPasa陽性細胞の検索を行なった。これらの研究結果からL細胞の動態についておよその情報が得られたものと考える。以下にL細胞の動態を総括すると:1)マウスでは胎生期に幼弱なL細胞が表皮に入る。この細胞はATPasa陽性であるがIa抗原を発現していない。2)細胞は生後1週間以内にIa抗原を発現し、バ-ベック顆粒を形成して形態的に成熟し、その数も成体のレベルに増加する。これは出生によって皮膚が免疫学的刺激を受けることと呼応する。3)これに続いて表皮からリンパ筋へL細胞が流入するようになる。細胞はリンパ管を経由してリンパ節の皮質深層に達し、そこでTリンパ球やID細胞と密着する。つまり、そこが免疫応答の主要な場であることがうかがえる。4)消化管上部粘膜のL細胞は出現が遅く、生後3週で増加を始める。これは哺乳終了期と呼応するとみられる。5)マウス前胃部ではL細胞が長い突起を伸ばして広い領域を占め、免疫学的監視に対応するとみられる。6)表皮・粘膜上皮ともに、L細胞はThyー1陽性細胞と密接な関係を持つ。7)鳥類の表皮にもL細胞に類似の細胞が発見された。
